The Optimization of Chloropromazine (CPZ) Treatment Condition for in vitro Phototoxicity Test

As cosmetic products become more and more essential in modern society and are applied daily all through our lives, the significance of the safety assessment of cosmetic ingredients cannot be overemphasized. Phototoxicity test is used to identify whether the phototoxicity is induced by the excited chemical after exposure to light and the alternative method generally used as a standard method is the in vitro 3T3 NRU phototoxicity test (OECD Test Guideline 432). Because there is no comment about the treatment concentration of positive control chloropromazine (CPZ) in the test guideline, we treated 500 ¥ìg/ml of CPZ as the maximum treatment concentration. In this condition, CPZ can be classified as a phototoxic material according to Photo Irritation Factor (PIF) criteria. However, sometimes IC50 value of CPZ didn¡¯t meet the standard criteria of acceptable ranges (CPZ irradiated (+Irr): IC50 = 0.1 to 2.0 ¥ìg/ml, CPZ non-irradiated (-Irr): IC50 = 7.0 to 90.0 ¥ìg/mL) due to the too high level of treatment concentration. As a result, the experiments often had to be performed repeatedly to get reliable data. To resolve this problem and increase the reliability of experimental data, we investigated the optimal CPZ concentration of treatment. The results showed that the maximum treatment concentration of 125 ¥ìg/ml and dilution factor (DF) of 3 made the experimental data most reliable and reproducible.
